寻常性银屑病患者外周血白介素27、白介素35及EBI3 mRNA表达的研究

Expressions of interleukin-27, -35 and Epstein-Barr virus-induced gene 3 mRNA in peripheral blood of patients with psoriasis vulgaris

摘要： 目的 探讨不同病期寻常性银屑病患者外周血白介素２７（IL-27）、白介素３５（IL-35）及其共同亚基EBI3 mRNA的表达。方法 分别采集寻常性银屑病进展期（22例）、静止期（20例）、消退期（20例）患者及30例健康对照外周血，采用竞争性免疫抑制ELISA法检测血清IL-27、IL-35表达水平，用SYBR Green实时荧光定量RT-PCR方法检测外周血单一核细胞中EBI3 mRNA的表达水平。 结果 寻常性银屑病患者血清内IL-27和IL-35表达均高于健康对照组。进展期、静止期、消退期IL-27（226.72 ± 71.32 ng/L，160.80 ± 20.02 ng/L，181.93 ± 27.36 ng/L）表达与健康对照组（138.66 ± 33.93 ng/L）相比，差异均有统计学意义（P值分别 < 0.01，< 0.05，< 0.01）；进展期、消退期血清IL-27表达显著高于静止期，差异有统计学意义（P值分别 < 0.01，< 0.05）。消退期血清IL-35（355.78 ± 49.82 ng/L）表达水平显著高于进展期（309.36 ± 36.10 ng/L）、静止期（299.14 ± 31.98 ng/L）、健康对照组（292.43 ± 36.26 ng/L），差异均有统计学意义（P值分别 < 0.01，< 0.01，< 0.01），进展期、静止期与健康对照组间比较，差异无统计学意义。外周血单一核细胞EBI3 mRNA在寻常性银屑病患者进展期、静止期、消退期表达量较健康对照组分别高20.15、16.17、19.74倍。 结论 IL-27、IL-35、EBI3作为抗炎因子可能参与寻常性银屑病发病。

Methods Peripheral blood was collected from patients with psoriasis vulgaris （PV） in progressive （n = 22）, stationary （n = 20） and regressive （n = 20） stage as well as 30 healthy human controls. A competitive enzyme-linked immunosorbent assay （ELISA） was performed to determine the serum level of IL-27 and IL-35. SYBR Green-based real-time reverse transcription （RT）-PCR was carried out to measure the mRNA expression of EBI3 in peripheral blood mononuclear cells （PBMCs）． Results The patients with psoriasis vulgaris showed a higher level of IL-27 and IL-35 compared with the healthy controls. The serum level of IL-27 was significantly higher in the patients with progressive, stationary and regressive psoriasis vulgaris than in the healthy human controls （（226.72 ± 71.32） ng/L, （160.80 ± 20.02） ng/L and （181.93 ± 27.36） ng/L vs. （138.66 ± 33.93） ng/L, P < 0.01, 0.05, 0.01, respectively）, and higher in the patients with progressive and regressive psoriasis vulgaris than in those with stationary psoriasis vulgaris （P < 0.01, 0.05, respectively）. Increased serum level of IL-35 was observed in the patients with regressive psoriasis compared with those with progressive and stationary psoriasis and healthy controls （（355.78 ± 49.82） ng/L vs. （309.36 ± 36.10） ng/L, （299.14 ± 31.98） ng/L and （292.43 ± 36.26） ng/L, all P < 0.01）, while no statistical difference was observed between the patients with progressive and stationary psoriasos and healthy controls. The mRNA expression level of EBI3 in the patients with progressive, stationary and regressive psoriasis was 20.15, 16.17 and 19.74 times, respectively, higher than that in the healthy controls. Conclusion IL-27, IL-35 and EBI3 may function as anti-inflammatory factors in the development, as well as the reversal of progression, of psoriasis. 【Key words】 Psoriasis; Interleukin-27；Interleukin-35；EBI3 mRNA
