羟基磷灰石纳米粒子抑制皮肤鳞状细胞癌细胞增殖的机制

Inhibition mechanism of hydroxyapatite nanoparticles on skin squamous cell carcinoma cells

摘要: 目的:探讨羟基磷灰石(HAP)纳米溶胶抑制皮肤鳞癌细胞株(A431)增殖的作用机制。方法:实验组用含浓度为400mg/L HAP纳米溶胶的DMEM培养液培养A431细胞。在复合培养的第1、2、3、4、5天,通过免疫荧光显微镜观察A431细胞凋亡过程中形态结构的变化;透射电镜观察A431细胞超微结构的改变。结果:免疫荧光显微镜下显示,实验组在复合培养的第1、2天出现少量绿荧光阳性细胞,细胞形态不规则,第3天可见红绿荧光双标阳性细胞,第4、5天出现较多的红荧光阳性细胞,细胞的染色质聚集于核膜周围成块状或新月形;透射电镜下可见实验组在复合培养的第2天细胞内出现HAP纳米粒子,细胞形态各异,胞质空泡化,核仁裂解,染色质边集,部分细胞胞质脱落。结论:HAP纳米溶胶抑制A431细胞增殖的作用机制可能为:HAP纳米粒子进入细胞内,直接作用于细胞内的细胞器,引起细胞凋亡、胀亡和胞体割裂。

Abstract: Objective: Discussing the inhibition mechanism on the nanometer hydroxyapatite solution (the nm-HAP-sol) on the skin squamous cell carcinoma cell line (A431). Methods: A431 cells were cultured with DMEM and the corresponding concentration of nm-HAP-sol in the 400mg/L. At the 1, 2, 3, 4, 5 days, observing the change of A431 cell morphosis during apoptosis through immunofluorescence microscopy and the variation of cell ultrastructure by transmission electron microscope. Result: It showed that there were few green fluor positive cells with irregular appearance at the 1, 2 days, double positive cells with green and red fluor emerged at the 3 day, there were many red fluor positive cells with crescent-shaped or massive caryotin assembling under the caryotheca at the 4, 5 days in experimental group through immunofluorescence microscopy. HAP nanoparticles appear at the 2 day of culture cells in experimental group by transmission electron microscope. It can be seen that cell morphology varied, cytoplasmic vacuolization, nucleolar cleavage, chromatin margination and partly cytoplasmic shedding. Conclusion: The HAP nanoparticles inhibited the generation of A431 cell by entering into the cell and effecting on the organelle and causing the generating cell apoptosis, oncosis and autoschizis.
