缺氧诱导因子1α和葡萄糖转运蛋白1在银屑病皮损中的表达

Expressions of hypoxia-inducible factor 1α and glucose transporter type 1 in psoriatic lesions

摘要:

目的 探讨缺氧诱导因子1α(HIF-1α)和葡萄糖转运蛋白1(GLUT-1)在银屑病皮损中的表达及其与角质形成细胞增殖的关系。方法 应用免疫组化技术和蛋白印迹对30例寻常性银屑病皮损、20例正常人皮肤组织中HIF-1α和GLUT-1的水平进行检测。结果 正常人皮肤组织中HIF-1α、GLUT-1主要表达在基底层,而银屑病中HIF-1α、GLUT-1均为全层表达;与正常人对照相比,银屑病皮损中HIF-1α和GLUT-1表达明显上调(P < 0.01),并与Ki-67表达呈正相关(r值分别为0.70和0.81,P均 < 0.01);GLUT-1的表达与HIF-1α的表达呈正相关(r = 0.85,P < 0.01)。结论 HIF-1α和GLUT-1在银屑病皮损中表达上调可能利于角质形成细胞的增殖,参与银屑病的发病。

Abstract:

Objective To investigate the expression of glucose transporter type 1(GLUT-1) and hypoxia-inducible factor 1 (HIF-1) α in psoriatic lesions, and to explore their correlations with keratinocyte proliferation. Methods Biopsy specimens were obtained from 30 patients with psoriasis and 20 normal human controls. Immunohistochemistry and Western blotting were used to examine the protein expression of GLUT-1 and HIF-1α in these specimens. Results GLUT-1 and HIF-1α were mainly expressed in the basal layer of the control skin, but throughout the whole epidermis of psoriatic lesions. A significant increase was observed in the expression of GLUT-1 and HIF-1α in psoriatic lesions compared with that in the control skin (both P < 0.01). In the case of psoriatic lesions, both the expression of GLUT-1 and HIF-1α was positively correlated with that of Ki-67 (r = 0.70, 0.81 respectively, both P < 0.01), and positive correlation was also found between the expression of GLUT-1 and HIF-1α (r = 0.85, P < 0.01). Conclusion Our data suggest that upregulation of GLUT-1 and HIF-1α expression in psoriatic lesions might contribute to the proliferation of keratinocytes and psoriasis development.
