系统性红斑狼疮患者淋巴细胞凋亡的研究

Study on Lymphocyte Apoptosis in Patients with Systemic Lupus Erythematosus

摘要: 目的 研究系统性红斑狼疮患者 (SLE)淋巴细胞凋亡情况以及自身抗体产生的关系。方法 通过分离出淋巴细胞,在12孔板内培养0、24、48小时,用吖啶橙染色,于荧光显微镜下观察计数。结果 22例SLE患者的淋巴细胞凋亡率较16例正常对照组在培养24、48小时时有显著增高 (P均<0.01)。SLE患者伴有2种以上自身抗体阳性者,培养24、48小时具有更高的淋巴细胞凋亡率 (P<0.05和P<0.01)。结论 SLE患者淋巴细胞在体外凋亡加速,可能与体内自身抗体产生增多有关,其淋巴细胞凋亡异常在其发病机制中起一定作用。

Abstract: Objective To assess the rate of apoptosis of lymphocytes isolated from patients with systemic lupus erythematosus(SLE) and investigate the correlation between lymphocyte apoptosis(LA) and autoantibody production. Methods Lymphocytes were isolated by Ficoll-Hypaque gradient,cultured in 12-well plates for 0,24,48 hours,and examined by acridine orange(AO) staining under fluorescence mi-croscope.Results The rates of LA of 22 SLE patients were sigificantly increased at t24(P<0.01) and t 48(P<0.01) com pared with those of 16 normal controls. The more severe the disease activity,the higher the rate of LA.In the patients with≥2 autoantibodies higher levels of LA at t24(P<0.05) and t48(P<0.01) were observed. Conclusion The results indicate that the in vitro acceleration of LA in SLE patients may be related to the increased production of autoantibodies,and the abnormalities of LA may play a role in the pathogenesis of SLE.
