寻常型银屑病皮损TNFα、核因子κB及诱生型NO合成酶表达

Expression of Tumor Necrosis Factor α, Nuclear Factor κB and Inducible Nitric Oxide Synthetase in Lesional Skin of Psoriasis Vulgaris

摘要: 目的 探讨诱生型NO合成酶(iNOS)在银屑病皮损中表达的调节机制.方法 应用免疫组化技术对寻常型银屑病皮损中肿瘤坏死因子(TNFα)、核因子(NF)κB及iNOS表达进行了检测.结果 15例正常人皮肤均不表达NFκB,5例于毛囊、汗腺、皮脂腺或立毛肌处表达TNFα,9例表皮全层较弱且均匀表达iNOS,5例真皮内单一核细胞(MNCs)弱表达iNOS.26例寻常型银屑病皮损表皮角质形成细胞及真皮内MNCs均表达TN-Fα、NFκB及iNOS,在Munro微脓疡处表达较强.角质形成细胞及真皮内MNCs上TNFα与iNOS表达强度呈显着正相关(rs'分别为0.526和0.665,P分别小于0.01和0.001),而NFκB与TNFα和iNOS之间无明显相关(P均>0.05).结论TNFα以NFκB非依赖性机制诱导寻常型银屑病皮损表皮角质形成细胞及真皮内MNSs表达iNOS,而在Munro微脓疡处则可能以NFκB依赖性机制诱导中性粒细胞表达iNOS.

Abstract: Objective To explore the regulatory mechanism of inducible nitric oxide synthetase (iNOS) expression in lesional skin of psoriasis. Methods The expression of tumor necrosis factor α(TNFα), nuclear factor κB (NF κB) and iNOS was detected by immunohistochemical technique. Results Fifteen skin samples of healthy controls did not express NF κB, 5 of them expressed TNFαin hair follicles, sweat glands, sebaceous glands or arrectores pilorum, 9 of them slightly and evenly expressed iNOS in all layers of epidermis, 5 of them also slightly expressed iNOS on mononuclear cells (MNCs) in dermis. The TNFα, NF κB and iNOS were expressed on epidermal keratinocytes and dermal MNCs, and strongly expressed in Munro's microabscess in lesional skin of 26 cases of psoriasis vulgaris. The intensity of expression of TNFαand iNOS was significantly correlated between keratinocytes and dermal MNCs (rs'were 0.526 and 0.665, respectively, P<0.01 and 0.001, respectively), and the intensity of expression of NF κB was not correlated to TNFα or iNOS between two kinds of cells in psoriasis vulgaris. Conclusion TNFα induces iNOS expression on keratinocytes and MNCs in lesional skin of psoriasis vulgaris through NF κB-independent mechanism, and on neutrophils in Munro's microabscess through NF κB-dependent mechanism.
