金黄色葡萄球菌肠毒素A诱导黑素瘤B16细胞凋亡的体外研究

in vitro induction of apoptosis in murine B16 melanoma cells by staphylococcus enterotoxin A

摘要: 目的 探讨超抗原金黄色葡萄球菌肠毒素A(SEA)诱导黑素瘤B16细胞凋亡.方法 用不同浓度的SEA(0.001、0.01、0.1、1 μg/mL)作用于体外培养的B16细胞,采用荧光法观察SEA诱导B16细胞凋亡的形态学改变,用末端标记法(TUNEL)计算细胞凋亡率,用流式细胞仪分析B16细胞的凋亡周期.结果 三种方法均显示SEA对小鼠B16细胞有明显的诱导凋亡作用,SEA在0.01 μg/mL浓度时B16细胞的凋亡率最高.结论 超搞原SEA对体外培养的小鼠B16细胞有诱导凋亡的作用.

Abstract: Objective To investigate the in vitro induction of apoptosis in marine B16 melanoma cells by staphylococcus enterotoxin A(SEA).Methods Marine B 16 melanoma cells were cultured in vitro and treated with different concentrations(0.001,0.01,0.1,1 wg/mL) of SEA.A fluorometric method was used to observe the morphological changes of cell apoptosis.Cell apoptosis rate was calculated by terminal transferase dUTP nick end labeling(TUNEL) assay.Flow cytometry was applied to detect the changes of cell cycles.Results It was shown that the apoptosis of B16 cells could be induced by superantigen SEA.And the highest apoptosis rate resulted from the inducement by 0.01 wg/mL of SEA.Conclusion Superantigen SEA could induce the apoptosis of marine B16 melanoma cells in vitro.
