中波紫外线影响大鼠表皮角朊细胞p53蛋白及p21蛋白表达的研究

Coexpression of p53 and p21 in Rat Epidermal Keratinocytes Following UVB Irradiation

摘要: 目的 探讨表皮角朊细胞对抗中波紫外线(UVB)损伤和致癌的机制 方法 采用LSAB免疫组化方法, 测定UVR照射后大鼠正常表皮p53蛋白及p21蛋白表达.结果 UVB单次照射后4小时即能测到p53蛋白表达, 第8小时左右达到峰值, 72小时后消失;p21蛋白出现时间与p53蛋白相同, 但其消退较慢, 第72小时仍能被检测到.两者染色主要位于表皮角朊细胞核内.结论 表皮角朊细胞可能是通过野生型p53蛋白和p21蛋白来对UVB导致表皮角朊细胞癌变起抑制作用.

Abstract: Objective To investigate the function of p53 and p21 in epidermal keratinocytes following UVB irradiation.Methods Using immunohistoehemical staining, the appearance of p53 protein and p21 was detected in the epidermis of rat after exposure to UVB in'adiation.Results The positive reactivity to p53 protein was found at 4h rollowing UVB irradiation, peaked at 8h and returned to undeteetable level at 72h. The appearance of p21 protein was also observed at 4h, but disappeared a little later, and eouht still be detected at 72h.Conclusion UVB caused a rise in wild-type p53 protein levels, in association with induction of p21 in rat epidermal keratinneytes.The above data suggest that over-expression of wild-type p53 and p21 may play an important role in preventing the carcinogenesis from damage of UVB.
