银屑病抗体搭桥角朊细胞/单一核细胞粘连:白介素2及白介素6和白介素2受体α链的检测

Psoriatic Antibody Bridging Keratinocyte/Mononuclear Leucocyte Adhesion: Estimation of IL-2, IL-6 and IL-2Rα

摘要: 抗体搭桥角朊细胞/单一核细胞粘连(ABKMA)作为银屑病研究一个体外实验模型,用来研究银屑病的免疫激活的细胞因子网改变。正常人表皮细胞悬液分离自包皮,加入银屑病IgG孵育后加入银屑病单一核细胞,形成ABKMA.使用ABC增强的ELISA检测培养上清的白介素2和白介素6.实验结果表明,白介素2和白介素6的合成和分泌都高于正常人IgG对照和无血清培养基对照。细胞表面的白介素2受体α链(IL2Rα)用免疫组织化学染色法显示。IL2Rα+单一核细胞/单一核细胞为4.6%.IL2Rα单一核细胞/粘连的单一核细胞为40.4%,这两者都高于相应的对照。

Abstract: Antibody bridging keratinocyte/mononuclear leucocyte adhesion(ABKMA), an experimental model in vitro for psoriasis, has been used in the investigation of cytokine network changes. ABKMA was formed by using normal human epidermal cells incubated with IgG of patients with psoriasis, and then mononuclear leucocytes(ML) of patients with psoriasis were added. Interleukin 2 and interleukin 6 in culture supernate were estimated by ABC amplified ELISA. The results indicated that the synthesis and secretion of interleukin 2 and interleukin 6 were higher than normal IgG and serum free medium controls. Immunohistochemistry staining was used to demonstrate the cell surface interleukin 2 receptor α chain (IL2Rα). The expression of IL2Rα+ML/ML and IL2Rα+ML/adhesion ML was 4.6% and 40.4% respectively. Both these data were higher than corresponding controls.
