Molecular identification of Trichophyton rubrum strains isolated from multiple body sites of one patient

Abstract: Objective We report a case of patient with six-lesion of Trichophytosis, the isolate from each lesion was identified to species and strain level, and their relationship was analyzed by means of the morphological, biochemical and molecular biological methods. Methods Primarily, the 6 strains were identified by morphological and biochemical methods, then the internal transcribed spacer (ITS) products was sequenced after PCR, the results indicated that all of isolates were Trichophyton rubrum. Then T.rubrum specialized RAPD and Trs-1/Trs-2 amplification were conducted to differentiate the 6 strains. Results There are 5 different strain types according to RAPD while 3 different strain types in Trs-1 result, which could differentiate the strains that RAPD could not does. Conclusions All of the six-lesions were infected different strains of T. rubrum by combination of RAPD, Trs-1/Trs-2 PCR application, indicating that the multiple origins and colonies of the same one patient with Trichophytosis rubrum.
