CpG containing oligodeoxynucleotides enhance the induction of IgG subtype of anti-dsDNA antibodies by kinetoplast DNA derived from Trypanosoma equiperdum

Abstract: Objective To study the role of CpG containing oligodeoxynucleotide(ODN)in the in-duction of IgG subtype of anti-dsDNA antibodies by kinetoplast DNA(kDNA)derived from Try-panosoma equiperdum(TE).Methods kDNA was extracted from TE and purified,then was injected sub-cutaneously into immunize normal BALB/c mice,with CpG ODN,incomplete Freund's adjuvant(IFA),or a mixture of CpG ODN and IFA as immune adjuvant.The serum titers of anti-dsDNA antibodies were deter-mined with TE as the substrate by ELISA after immunization.The levels of total IgG,C3,IL-10 and IFN-γwere also detected by single immunodiffusion test and ELISA respectively.Results Without any immune adjuvant,TE kDNA alone could only induce IgM subtype of anti-dsDNA antibodies,while CpG ODN or I-FA induced the IgG subtype of anti-dsDNA antibodies.CpG ODN had a stronger capability to enhance the induction of anti-dsDNA antibodies by kDNA than IFA(P<0.05).Conclusion CpG ODN can enhance the induction of IgG subtype of anti-dsDNA antibodies by TE kDNA efficiently.
