Effect of octreotide on the secretion of cytokines by and proliferation of cultured PBMCs from patients with systemic lupus erythematosus

Abstract: Objective To investigate the effect of octreotide (OCT) on the proliferation capacity of and secretion of cytokines by cultured PBMCs from patients with systemic lupus erythematosus (SLE). Methods PBMCs from patients with SLE and healthy controls were cultured in vitro with various concentrations of OCT,pretreated with or without growth hormone (GH).The proliferation capacity of PBMCs was detected by MTT method,and the secretion of cytokines by PBMCs was measured with ELISA.Results OCT could inhibit the proliferation capacity of PBMCs in a dose dependent manner:0.1 ng/mL of OCT had no effect on the proliferation,1 ng/mL OCT had a weak inhibitory effect,and 10 ng/mL OCT had a marked inhibitory effect.The secretion of IL-6,IL-10 and IFN-γby PBMCs was significantly decreased with the treatment of OCT,and was also significantly decreased with the co-treatment of GH and OCT compared to that with the treatment of GH only.Conclusion OCT can inhibit the proliferation capacity of PBMCs and secreation of IL-6,IL-10 and IFN-γby PBMCs induced by GH.
