环氧合酶2在人恶性黑素瘤中高表达

Overexpression of cyclooxygenase-2 in human malignant melanoma

摘要: 目的 探讨恶性黑素瘤组织及黑素瘤细胞系中环氧合酶2(COX-2)的表达及意义.方法 免疫组化法比较性研究25例皮内痣、45例恶性黑素瘤COX-2蛋白的表达情况.并结合患者的临床资料进行分析.同时,免疫印迹法检测正常黑素细胞及3种黑素瘤细胞系中COX-2蛋白的表达情况.结果 在恶性黑素瘤中,COX-2蛋白阳性表达率为80%(36/45),显著高于皮内痣组0%(0/25)(P<0.05);肿瘤组织中COX-2蛋白表达与淋巴结转移、TNM分期密切相关(P<0.05).正常黑素细胞COX-2阴性表达,3种恶性黑素瘤细胞系中A375、1585细胞COX-2表达水平明显升高,而Libr细胞未见阳性表达.结论 COX-2在恶性黑素瘤组织及细胞系中表达率显著升高,可能在恶性黑素瘤发生发展中具有一定作用.

Abstract: Objective To investigate the expression of cyclooxygenase-2 (COX-2) in malignant melanoma.Methods The expression of COX-2 was determined by immunohistochemistry in 45 cases of malignant melanoma and 25 intradermal nevi.The correlation between the immunohistochemistry findings and clinical data (including age,gender,TNM staging,and presence of lymph node metastases) was also analyzed.The COX-2 expression in three human melanoma cell lines (A375,CRL-1585,and Libr) and cultured human melanocytes was assessed by Western blot.Results Thirty six of the 45 malignant melanoma samples and none of the 25 intradermal nevi were positive for COX-2.The positivity rates of COX-2 in lymph node metastasis group and in advanced stage (TNM stagesⅢandⅣ) were significant higher than those in lymph node negative group and in early stage (TNM stagesⅠandⅡ)(92.3% versus 63.2%,89.7% versus 62.5%,respectively;P<0.05).The expression of COX-2 was significantly associated with the ectopic expression of β-catenin (r=0.45).Western blot analysis confirmed negative COX-2 expression in normal human melanocytes and Libr melanoma cell line,whereas the expression of COX-2 was markedly elevated in A375 and CRL-1585 melanoma cells.Conclusions The results indicate that COX-2 is overexpressed in human malignant melanoma and melanocytes,and may be involved in the development and progress of melanoma.
