6味中药乙醇提取物对B16黑素瘤细胞酪氨酸酶及其相关蛋白-1、-2 mRNA表达的影响

Effects of ethanol extracts from six traditional Chinese medicines on the expression of mRNA for tyrosinase,tyrosinase-related protein-1 and-2 in B16 melanoma cells

摘要: 目的 研究墨旱莲(旱莲草)等6味用于治疗白癜风中药的乙醇提取物对小鼠B16黑素瘤细胞酪氨酸酶及酪氨酸酶相关蛋白-1、-2(TRP-1、-2)mRNA表达的影响.方法 半定量逆转录-聚合酶链反应(RT-PCR)测定酪氨酸酶及TRP-1、-2 mRNA的表达,并与阳性对照甲氧沙林(8-MOP)进行比较.结果 山慈姑(毛慈姑)、苦参、黑芝麻和墨旱莲的乙醇提取物显著上调细胞酪氨酸酶mRNA的表达(P<0.05);除苦参对TRP-2 mRNA有明显上调作用外(P<0.05),其他中药对TRP-1和TRP-2 mRNA的表达无明显作用(P>0.05).结论 山慈姑、苦参、黑芝麻和墨旱莲有上调酪氨酸酶mRNA的作用,苦参还可上调TRP-2 mRNA的表达.

Abstract: Objective To study the effects of ethanol extracts from six traditional Chinese medicines for treating vitiligo on the expression of mRNA for tyrosinase (TYR),tyrosinase-related protein (TRP)-1,and TRP-2 in B 16 melanoma cells.Methods Ethanol extracts were prepared from six traditional Chinese medicines,i.e.Cremastra appendiculata (D.Don) Makino,Mentha haplocalyx Briq.,Sophora flavescens Ait.,Prunella vulgaris L.,Sesamum indicum L.,and Eclipta prostrata L..B16 melanoma cells were treated respectively with ethanol extracts at a concentration of 10 or 20μg/mL for 72 h,with culture media and 8-methoxypsoralen (8-MOP) served as blank and positive control,respectively.The expression of mRNA for TYR,TRP-1,and TRP-2 was measured by semiquantitative reverse transcription PCR.Results The ethanol extracts from Sophora flavescens Ait.significantly upregulated the expression of mRNA for both TYR and TRP-2 (both P<0.05).The expression of TYR mRNA was also significantly upregulated by extracts from Cremastra appendiculata (D.Don) Makino,Sesamum indicum L.,and Eclipta prostrata L. (all P<0.05),whereas none of the extracts affected TRP-1 mRNA expression (P>0.05).Conclusions The results indicate that Cremastra appendiculata (D.Don) Makino,Sophora flavescens Ait.,Sesamum indicum L.and Eclipta prostrata L.can upregulate the expression of TYR mRNA.Moreover,Sophora flavescens Ait can also upregulate TPR-2 mRNA expression.These may partially account for the therapeutic mechanisms of these Chinese herbal drugs in vitiligo.
