12株马内菲青霉随机扩增DNA多态性研究

DNA Polymorphisium of Penicillium marneffei by Random Amplification PCR Assay

摘要: 目的 了解马内菲青霉的基因学特征,为该菌的DNA分型提供依据。方法 采用氯化苄提取法提取基因组DNA.用随机扩增多态性DNA(RAPD)技术对12株临床分离的马内菲青霉菌株进行DNA指纹图谱分析。结果 ①共选用40个随机引物进行扩增,筛选出4个具有稳定、清晰DNA扩增带型的引物。即引物P23:5'-AGTCAGCCAC-3',P103:5'-AGACGTCCAC-3',P105:5'-AGTCGTCCCC-3',P120:5'-GGGAGACATC-3'.②12株菌株DNA图谱主要带型相同,扩增片段呈多态性。相似率44.4%~97.3%.③12株菌可分5个不同的组群。结论 RAPD技术可以用于马内菲青霉的基因分型。

Abstract: Objective To investigate the DNApolymorphism of Penicillium marneffei(PM)and to provide an evidence for PM classification.Methods Total DNA was extracted with benzyl chloride.Random amplification polymorphic DNA(RAPD)assay was used to study the finger-printing of 12 strains Penicillium marneffei.Results ①Of forty randomprimers used,four of them:P23:5'-AGTCAGCCAC-3',P103:5'-AGACGTCCAC-3',P105:5'-AGTCGTCCCC-3',P120:5'-GGGAGACATC-3' gave good reactio n.②The major bands of DNA patterns of the 12 strains were of the same mobility.The amplified bands were polymorphic.The similarities were in the range of 44.4%-97.3%.③12 strains could be divided into 5 groups.Conclusion RAPD may be applied in PMDNA classification.
