聚合酶链反应与细胞培养法检测临床标本中的沙眼衣原体

Comparison of Polymerase Chain Reaction and Cell Culture for Detection of Chlamydia Trachomatis in Urogenital Specimens

摘要: 为了评价PCR对泌尿生殖道标本中沙眼衣原体的检测结果,利用传统的细胞培养法进行了平行检测比较.泌尿生殖道标本经裂解液(主要成分为非离子去污剂)裂解10分钟后,未经提纯DNA而直接用于PCR扩增.每份标本同时进行细胞培养.对PCR与培养结果不相符合的标本,取标本沉淀经直接免疫荧光(DFA)染色后进行确证.检测临床标本305份,其中有24份标本PCR阳性而培养阴性,经直接免疫荧光镜检证实为真阳性.另有5份培养结果阳性的标本未能被PCR检出.综合PCR、培养及DFA的结果,共检出阳性93例,阴性212例.以该结果作为“扩大的金标准”,PCR的敏感性为94.62%(88/93),特异性为100%(212/212),培养法的敏感性为74.19%(69/93),特异性为100%(212/212).结果表明PCR是一高度特异和敏感的检测方法.

Abstract: A study to compare the polymerase chain reaction(PCR) test with the cell culture method in detection of Chlamydia trachomatis in urogenital specimens infections was performed.Urogenital specimens were simply lysed by nonionic detergents for 10 minutes and subsequently subjected to amplification without prior extraction of nucleic acid.
