Association between the polymorphism in interleukin-10 gene promoter and systematic lupus erythematosus in a Chinese Zhuang population in Guangxi Zhuang Nationality Autonomous Region

Abstract: Objective To investigate the association between the polymorphism of interleukin -10 (IL -10) gene promoter and systematic lupus erythematosus(SLE)in a Chinese Zhuang population in Guangxi Zhuang Nationality Autonomous Region.Methods Blood samples were collected from 90 patients with SLE and 110 age-and sex-matched normal human controls in a Chinese Zhuang population.PCRrestriction fragment length polymorphism(PCR-RFLP) was performed to assess the single nucleotide polymorphisms(SNPs) at positions -1082,-819 and -592 of IL -10 gene promoter.Results There was no significant difference in either genotype distribution or allele frequency at positions-819 and-592 between the SLE patients and normal controls(both P>0.05).However,statistical difference was observed in the genotype frequency of -1082(A/G) in IL -10 gene between the two groups.The relative risk analysis of genotype frequency revealed that the risk for SLE in carriers of genotype -1082 AG was 2.076 times of that in carriers of -1082 AA(OR=2.076,95% CI:1.052~4.100).The haplotype GCC(G at position -1082, C at position -819 and C at position -592) significantly increased the risk for SLE.Conclusion The polymorphism at position -1082 and genotype of IL -10 gene promoter may be associated with the occurrence of SLE,and -1082 G allele may confer risk for SLE.
