Rapid identification of five common species of Candida with real-time fluorescence-based quantitative PCR

Abstract:

[Abstract] Objective: To establish a Real-time quantitative PCR way in detection and identification of Candida species and explore its feasibility. Method:fungal universal primer ITS4 and ITS86 were designed. Five Candida species were applied to construct recombinant plasmids. Recorded Melting temperature curves after finishing detection by real-time quantitative PCR. Analyzed data. Results: Melting temperatures of five Candida species: (87.33±0.13)℃(Candida albicans), (84.39±0.20)℃(Candida tropicalis), (85.53±0.18)℃(Candida parapsilosis), (86.47±0.10)℃(Candida glabrata), (91.41±0.18)℃(Candida krusei). Conclusion: This study shows the method of Real-time quantitative PCR is a rapid way in detection and identification of Candida species.
