Effects of all-trans-retinoic acid andγ-interferon on the expression of survivin in SCL-1 cells

Abstract: Objective To study the effects of all-trans-retinoic acid(ATRA)andγ-interferon(γ-IFN)on the expression of survivin in skin squamous cell carcinoma cell line SCL-1.Methods SCL-1 cells were treated withγ-IFN alone(100,500,1000 u/mL,respectively)or in combination with ATRA(1000 u/mLγ-IFN+10-6mol/mL ATRA)in vitro.The mRNA and protein expressions of survivin were detected in SCL-1 cells by RT-PCR and Western-blot respectively.Result sγ-IFN could significantly inhibit the mRNA and protein expressions of survivin in a dose-and time-dependent manner.Also,ATRA could enhance the inhibition of survivin expression byγ-IFN.Conclusion The down-regulation of survivin ex-pression may be a mechanism for the induction of apoptosis of skin squamous cell carcinoma cells by the combination ofγ-IFN and ATRA.
