Enhancement of human papillomavirus type 11 DNA vaccine-induced specific immune response by co-immunization with CD80 co-stimulatory gene

Abstract: Objective To investigate the specific immune response induced by DNA vaccinal plasmid of human papillomavirus type 11 (HPV11)E7,L1-E7 and enhancement of immune reaction by CD80 co-stimulatory gene.Methods The constructed pcDNA3.1 (+)/HPV11E7,E7-CD80 and L1-E7 DNA vaccinal plasmid,or combination of CD80 and L1-E7 plasmids,were injected intramuscularly into mice,respectively.The distribution of plasmid DNA and transcription of RNA in tissue were analyzed by PCR and RT-PCR,respectively.The lymphocyte proliferation activity was detected by MTT assay.The level of cytokines was determined by ELISA in supernatants of cultured spleen cells and antibodies in serum.Results After immunization,the recombinant plasmid was mainly located in the injection site.Specific anti-HPV 11 E7 IgG/L1 IgG antibodies,lymphocyte proliferation and secretions of cytokines IL-2 and IFN-γwere induced by injection of DNA vaccine.The levels of lymphocyte proliferation and cytokine secretion were significantly higher in groups of HPV11E7 in recombinant with CD80 co-stimulatory gene than those without CD80.The levels of specific cellular and humoral immunity were also significantly increased in group with the combination of L1-E7 plasmid and CD 80 than those without CD80.Conclusions HPV11DNA vaccination could induce specific cellular and humoral immunity.CD80 co-stimulatory gene may represent an attractive adjuvant for enhancement of the specific immune responses.
