Characterization of Cladosporium carrionii and Cladophialophora ajelloi by Random Amplification of Polymorphic DNA Assay

Abstract: In order to investigate the DNA polymorphism of Cladosporium carrionii and Cladophialophora ajelloi,and to find the relationship of DNA patterns with morphology and geographic origin,the total DNA was extracted by using benzyl bromide method,and random amplification of polymorphic DNA(RAPD) assay was used to the study of genetic similarity(GS) of 21 isolates from five countries(19 of Cladosporium carrionii,2 of Cladophialophora ajelloi).Results:(1)Of ten random primers used,three gave good reaction.(2) The DNA fragment patterns of 19 isolates of Cladosporium carrionii showed high genetic similarity,but a little difference in comparison with that of 2 isolates of Cladophialophora ajelloi.The RAPD patterns of two isolates showed high genetic similarity with most Chinese isolates of Cladosporium carrionii(GS:85%).(3) The genetic similarity of Cladosporium carrionii are closely related to the geographic origin,the DNA patterns of isolates from the same country showed high similarity and located in the same group of dendrogram.RAPD assay can be used for the study of epidemiology and genetic characterization as a simple,rapid method.
