双嘧达莫抗单纯疱疹病毒的体外实验研究

Study on Anti Herpes Simplex Virus Activity of Dipyridamole in Vitro

摘要: 目的和方法 用Vero细胞及HEK-293细胞体外培养法检测双嘧达莫对单纯疱疹病毒(HSV)的抑制活性,并以直线回归方程计算IC50值.结果 双嘧达莫对HSV-1及2型质控株及临床分离株的IC50值范围在69~161μg/ml之间,而对照药阿昔洛韦为19~213μg/ml.结论 从双嘧达莫广谱的抗病毒作用及其特有的作用机制以及药代动力学资料分析,口服给药对HSV感染的疗效尚待临床药理研究进一步证实,而局部给药则可能是抗HSV皮肤粘膜感染可供选择的治疗药物之一.

Abstract: Objective and Methods To determine the inhibitory activity of dipyridamole (DPM) on replication of herpes simplex virus (HSV)-Ⅰ、Ⅱ in Vero cells and HEK-293 cells in vitro. Results The results showed that the range of 50% inhibition concentrations (IC50) of DPM and control drug acyclovir on different types of HSV in 2 cell lines ranged 6.9~16.1μg/ml and 1.9~21.3μg/ml respectively. Conclusion It is known that DPM possessed a definite antiviral activity with a broad spectrum, the therapeutic effect of DPM taken orally for the treatment of viral diseases should be further studied. It is possible that topical preparations of DPM can be used for the treatment of mucocutaneous infection of HSV.
