恶性黑素瘤细胞株细胞间粘附分子1的改变及临床意义

The Expression of Intercellular Adhesion Molecule-1 in Human Malignant Melanoma Cell Lines and Its Clinical Significance

摘要: 目的 探讨细胞间粘附分子1(ICAM-1)在恶性黑素瘤细胞株的表达及临床意义。方法 用体外细胞培养方法、逆转录聚合酶链反应技术,观察恶性黑素瘤细胞株MM81在温热及某些细胞因子刺激下,细胞形态、细胞增殖能力及ICAM-1的基因水平变化。结果 温热、γ-干扰素、肿瘤坏死因子α可使细胞形态改变、细胞增殖能力下降、ICAM-1的表达增加,白介素2未能影响细胞形态及增殖能力,也未能使ICAM-1明显改变。结论 恶性黑素瘤细胞株ICAM-1的表达与细胞被破坏有关,同时为温热、γ-干扰素、肿瘤坏死因子-α作为治疗恶性黑素瘤的辅助手段提供了理论依据。

Abstract: Objective To investigate the expression of ICAM-1 in human malignant melanoma (MM)cell lines and its clinical significance.Methods Human MM cell lines MM8.1 were cultured and analyzed by reverse transcription polymerase chain reaction(RT-PCR), the cell shape,cell growth ability and ICAM-1 by mRNA expression under the stimulation of hyperthermia or cytokines were observed.Results Hyperthermia、γ-IFN and TNF-α could change cell shape,reduce cell growth ability and increase the expression of ICAM-1.IL-2 could not influence the cell shape and the expression of ICAM-1.Conclusion The expression of ICAM-1 in MM cell lines may be correlated with the cell destruction,which provides theoretical basis of supplementary therapy for MM with hyperthermia、IFNγ and TNFα.
