玫瑰糠疹患者单一核细胞转录因子T-bet mRNA及GATA-3 mRNA的表达

Expressions of T-bet and GATA-3 mRNA in peripheral blood mononuclear cells of patients with pityriasis rosea

摘要:

目的 探讨T-bet、GATA-3在玫瑰糠疹发病机制中的意义。方法 SYBR Green I实时荧光定量PCR技术检测17例玫瑰糠疹患者和20例正常人外周血单一核细胞中T-bet mRNA和GATA-3 mRNA的表达。结果 玫瑰糠疹患者外周血中T-bet mRNA与 GATA-3 mRNA Avg △Ct值分别为3.33 ± 0.94与5.22 ± 0.69,对照组分别为4.31 ± 1.11与4.36 ± 1.02。患者的T-bet mRNA表达高于对照组(P < 0.05),GATA-3表达低于对照组(P < 0.05)。结论 玫瑰糠疹患者外周血中T-bet/GATA-3升高,Th1优势表达,细胞免疫异常在玫瑰糠疹发病过程中可能具有重要作用。

Abstract:

Objective To investigate the significance of T-bet and GATA-3 in the pathogenesis of pityriasis rosea. Methods SYBR Green I real-time fluorescence quantitative PCR was performed to detect the expressions of T-bet and GATA-3 mRNA in peripheral blood mononuclear cells (PBMCs) from 17 patients with pityriasis rosea and 20 normal human controls. Results The average delta Ct value of T-bet mRNA and GATA-3 mRNA was 3.33 ± 0.94 and 5.22 ± 0.69 respectively in the patients, 4.31 ± 1.11 and 4.36 ± 1.02 respectively in the controls. There was a higher expression of T-bet mRNA but a lower expression of GATA-3 mRNA in the patients compared with the controls (both P < 0.05). Conclusions Patients with pityriasis rosea show an increased ratio of T-bet to GATA3 expression and predominant expression of Th1-type cytokines. Abnormal cellular immunity may play an important role in the pathogenesis of pityriasis rosea.
