白念珠菌临床株多药耐药蛋白基因表达与氟康唑耐药的关系

Relationship between the expression of multidrug resistant protein genes and fluconazole resistance in clinical isolates of Candida albieans

摘要: 目的 探讨临床分离白念珠菌氟康唑耐药株/敏感株多药耐药蛋白基因CDR1、CDR2、MDR1的表达情况。方法 试剂盒法抽提白念珠菌氟康唑耐药株/敏感株的总RNA后,逆转录成cDNA,采用实时定量PCR技术观察白念珠菌多药耐药蛋白基因的表达。结果 白念珠菌耐药株组CDR2基因的ΔCT值为7.52±2.53,敏感株组为9.28±3.15,两组比较,t=2.37,P<0.05,耐药株的表达量要高于敏感株。结论 白念珠菌CDR2基因的高表达与氟康唑耐药有关。

Abstract: Objective To explore the relationship between fluconazole resistance and expression of multidrug resistant protein genes,including CDR1,CDR2,MDRI.Methods The total RNA was extracted from fluconazole-resistant and-susceptible Candida albicans isolates,and cDNA was synthesized.The expression of CDR1,CDR2 and MDRl genes was then detected by quantitative real-time PCR.TheΔCT (threshold cycle) value was obtained by subtracting the CT value of 18S rRNA from that of the targeted gene.Results TheΔCT value of CDR2 was significantly lower in fluconazole-resistant isolates than in fluconazolesusceptible isolates (7.52±2.53 vs.9.28±3.15,t=2.367,P<0.05),indicating the expression of CDR2 gene was higher in the fluconazole-resistant isolates than in the fluconazole-susceptible isolates.Conclusion It is likely that the over-expression of CDR2 gene is relevant to fluconazole resistance in Candida albicans.
