32株孢子丝菌临床株两种基因分型方法比较

摘要: 目的 确定哪种孢子丝菌基因分型的方法分辨力更高,所分基因型与临床表现及地域分布相关性更好。方法 以32株不同地区及临床类型的孢子丝菌菌株为研究样本,分别进行mtDNA RFLP分型,基因组DNA RAPD分型和rDNA RFLP-Southern blotting杂交分型,并进行比较。结果 mtDNA-RFLP方法将受试菌株分为5型,分别对应Ishizaki等命名的1,4,6,7,20五种基因型;RAPD方法将受试菌株分为7种基因型(I-VII型); rDNA RFLP-Southern blotting杂交方法将受试菌株分为15种基因型(A-O型),且其所分基因型与菌株的南北方分布及临床表现有一定相关性。结论 三种孢子丝菌基因分型方法中,rDNA RFLP-Southern blotting杂交方法分辨力较高,所分基因型与临床表现及地域分布相关性更好。

Abstract: Objectives To determine the best method of the genetic typing of Sporothrix schenckii. Methods The same samples (32 isolates of S.schenckii) were used to do mtDNA typing on RFLP pattern and DNA RAPD analysis and rDNA RFLP-Southern blotting analysisand then compared the three methods. Results 32 strains of S.schenckii were composed of 5 mtDNA types, respectively were type 1,4,6,7,20 (correspond to the work of Ishizaki et al).The DNA patterns could be divided into 7 types(typeI-VII) by RAPD analysis. 15 individual patterns (DNA type A-O) of all strains were recognized by the southern blotting,and had an obvious correlation with different geographic distribution and clinical manifestations. Conclution rDNA RFLP-Southern blotting assay for DNA typing of S.schenckii provides a highly sensitive.It is also found that there is an obvious correlation between DNA patterns and different geographic distribution and clinical manifestations by this assay.
