BAG-1蛋白在皮肤恶性黑素瘤皮损中的表达及其与P53、Bcl-2的关系

Overexpression of BAG-1 in human malignant melanoma and its association with p53 and bcl-2

摘要: 目的 探讨皮肤恶性黑素瘤组织及黑素瘤细胞株中BAG-1蛋白、P53、Bcl-2的表达及意义。方法 免疫组化法检测32例皮肤恶性黑素瘤、20例色素痣皮损石蜡包埋标本中BAG-1蛋白、P53和Bcl-2的表达情况,并结合患者临床资料进行分析;免疫细胞化学法检测BAG-1蛋白在黑素瘤细胞株M14和B16F10细胞的表达情况。结果 BAG-1蛋白在32例皮肤恶性黑素瘤组织中25例阳性,阳性表达率为78%;色素痣组20例中阳性3例,阳性表达率为15%,两组比较,差异有统计学意义(P < 0.001)。BAG-1蛋白在皮肤恶性黑素瘤组织中的表达与Clark分级呈正相关(r = 0.47,P < 0.05),与P53、Bcl-2表达无显著相关性(r 值分别为0.05和0.11,P > 0.05)。BAG-1蛋白在黑素瘤细胞株M14、B16F10细胞均呈阳性表达。结论 BAG-1蛋白在皮肤恶性黑素瘤组织中的表达显著升高,并与肿瘤的Clark分级呈正相关。

Abstract: Objective To investigate the expressions of BAG-1, P53 and Bcl-2 proteins and their significance in malignant melanoma. Methods Biopsy samples were obtained from 32 patients with malignant melanoma and 20 patients with pigmented nevi, and paraffin embedded sections were prepared. The expressions of BAG-1, P53 and Bcl-2 proteins were examined with immunohistochemistry in these sections. The correlation between the immunohistochemical findings and clinical data (including age, gender, Clark level and presence of lymph node metastases) was also analyzed. The expression of BAG-1 protein in melanoma cell lines, M14 and B16F10, was assessed by immunocytochemistry. Results The expression rate of BAG-1 protein was 78% (25/32) in cutaneous malignant melanoma, 15% (3/20) in pigmented nevi; the difference was statistically significant (P < 0.01). The expression of BAG-1 protein was positively correlated with Clark level of cutaneous malignant melanoma (r = 0.47, P < 0.05), but not with the expressions of P53 or Bcl-2 (r = 0.05, 0.11, respectively, both P > 0.05). Immunocytochemistry showed that BAG-1 protein was expressed in M14 and B16F10 cells. Conclusions The results indicate that BAG-1 protein is overexpressed in cutaneous malignant melanoma, and the overexpression is positively correlated with Clark level of malignant melanoma.
