单核细胞趋化蛋白-1及其受体在阴道念珠菌病小鼠模型中的表达

Expression of MCP-1 and its receptor CCR2 in a murine model of experimental vaginal candidiasis

摘要: 目的 探讨单核细胞趋化蛋白-1(MCP-1)及其受体CC趋化因子受体2(CC chemokine receptor 2.CCR2)在小鼠阴道念珠菌病发病中的作用.方法 建立阴道念珠菌病小鼠模型.以RT-PCR及ELISA方法检测小鼠阴道及腰淋巴结中MCP-1及CCR2的表达.结果 与正常小鼠相比,雌激素对MCP-1及CCR2的表达无明显影响.所有感染组小鼠阴道组织MCP-1的mRNA和蛋白水平均明显升高,其CCR2 mRNA水平亦相应升高;与之相反,所有感染组小鼠的引流腰淋巴结MCP-1的浓度与基础水平相比无明显改变,而雌激素处理感染组小鼠CCR2在接种后7~21d明显升高.结论 MCP-1及其受体CCR2可能在局部刚道黏膜白念珠菌感染中起一定的作用.

Abstract: Objective To investigate the role of monocyte chemoattractant protein-1(MCP-1) and its receptor CC chemokine receptor 2(CCR2)in vaginal infection with Candida albicans by detecting their expression in a murine model with experimental vaginal candidiasis.Methods A murine model of vaginal candidiasis was established.Mice infected with Candida albicans,and the non-infected control animals,were treated with estrogen.The expression of MCP-1 and CCR2 in the murine vaginal tissue and draining lumbar lymph nodes was assessed by RT-PCR and ELISA.Results Estrogen had no significant effect on the expression of MCP-1 or CCR2.Compared with the basal level of control mice,the protein and mRNA expression of MCP-1 as well as the CCR2 mRNA expression were significantly increased in the vaginal tissue of infected mice,while the protein and mRNA expression of MCP-1 in the draining lumbar lymph nodes of infected mice remained unchanged.A significant increase in the mRNA expression of CCR2 was observed in estrogen-treated infected mice at day 7,14 and 21 after the inoculation.Conclusion MCP-1 and its receptor CCR2 may play a role in the local vaginal mucosal response to C.albicans.
