犬小孢子菌入侵头发模式及其体外生长繁殖的观察

摘要: 目的 探讨犬小孢子菌入侵头发的过程、体外生长繁殖规律及不同发育阶段的形态特征.方法 取20例脓癣和白癣患儿断发、鳞屑标本,20%KOH制片,直接镜检;作病理切片PAS染色镜检;接种于沙氏培养基上培养分离原代菌落,再转种到米饭培养基上,25℃~28℃下培养,2d后观察.结果 直接镜检发现头屑内有菌丝,还有大小分生孢子.断发毛髓质外除有镶嵌式孢子以外,还有大分生孢子,或者自外向内依次为毛小皮层(护膜)、大分生孢子、小分生孢子;毛髓质内也有大分生孢子.病理切片显示,菌鞘部的病原菌形态与分布和断发残端及发根部完全不同,前者毛小皮层内侧有大量密集的圆形小分生孢子,而后者毛皮质和毛髓质内有大量菌丝及大小分生孢子.米饭培养基上,犬小孢子菌生长4d开始长出大分生孢子,其生长繁殖的规律是:分生孢子器→早期大分生孢子→成熟大分生孢子→大分生孢子内长出新的一代菌丝和分生孢子器→新生的大分生孢子继续生长发育,再产生新一代.结论 犬小孢子菌作为亲动物性皮肤癣菌,其侵人毛发各层内的菌丝和孢子破坏毛发组织,造成断发.犬小孢子菌在米饭培养基上大分生孢子的生长繁殖是以大分生孢子内产生新一代的菌丝和分生孢子器为特征.

Abstract: Objective To investigate the morphological characteristics during the invasion process in hair infection caused by Microsporum canis and its growth and proliferation in vitro.Methods Twenty children suffered from Microsporum canis-induced tinea alba or kerion were selected.The infected hairs and scales were taken for direct microscopic examination by 20% KOH preparation,fungal culture on Sabouraud's media and histopathological examination with PAS stain.Then the primary colonies were isolated,transinoculated to rice grain media and cultured at 25℃~28℃.The cultures were observed and photographed under a microscope 2 clays after the trans-inoculation.Results Micro-and macro-conidia as well as hyphae were found in the scales by the direct microscopic examination.In the infected hairs,there were mosaic spores with macroconidia outside the hair,or triple-layer structures with outer cell layer,macroconidia layer and microconidia layer sequentially from the exterior to the interior,and macroconidia inside the hair medulla.The histopathological examination showed different morphological features of fungi between the fungal sheath and the stump of broken hairs as well as hair roots.In the former,there was mosaic-like microconidia outside the hair while in the latter,large amount of hyphae and micro-as well as macro-conidia were inside the hair.The life cycle of Microsporum canis on the rice grain media presented a picture from conidia apparatus,early-stage macroconidia,mature macroconidia to new hyphae and pycnidia grown from macroconidia.New macroconidia continued to grow and new generation was produced,thus the fungi proliferated in cycles.Conclusions Microsporum canis is a zoophilic dermatophyte.The hyphae and spores invade into the hair tissue and cause the hair to break.The cycle of Microsporum canis on rice grain media is characterized by a new generation growth of hyphae and pycnidia from the macroconidia.
