系统性红斑狼疮患者B淋巴细胞刺激因子及其受体BAFF-R的表达

The expression of B lymphocyte stimulator and its receptor BAFF-R in PBMCs from patients with SLE

摘要: 目的 探讨系统性红斑狼疮(SLE)患者外周血单一核细胞(PBMC)中B淋巴细胞刺激因子(BLyS)及其受体BAFF-R(属于肿瘤坏死因子家族的B细胞活化因子受体)的表达情况.方法 采用半定量逆转录聚合酶链反应(RT-PCR)以及免疫印迹法(Westernblot)分别检测28例SLE活动期患者及24例SLE稳定期患者PBMC中BLyS与BAFF-R mRNA及蛋白的表达水平,并以21例健康志愿者作对照;同时将两者mRNA及蛋白的表达水平与SLE疾病活动程度进行相关分析.结果 SLE活动期、稳定期患者PBMC中BLyS与BAFF-R mRNA及蛋白的表达水平均明显高于正常人(P<0.01),活动期患者的表达高于稳定期患者(P<0.01);BLyS mRNA及蛋白的表达与狼疮活动指数(SLEDAI)呈显著正相关(P<0.01),BAFF-R mRNA及蛋白的表达与SLEDAI无明显相关性(P>0.05).结论 BLyS及其受体BAFF-R可能参与了SLE的发病机制.

Abstract: Objectives To investigate the expression of B lymphocyte stimulator (BLyS) and its receptor BAFF-R(a receptor of B-cell activator factor,belonging to the TNF family) in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE),and to explore their clinical significance.Methods The patients were separated into active group (n=28) and inactive group (n=24) according to the SLE disease activity index (SLEDAI).The mRNA and protein expression of BLyS and its receptor BAFF-R were measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western-blot in PBMCs from the patients and 21 healthy volunteers.The relationship between the expression of BLyS and BAFF-R and SLEDAI was analyzed.Results In the case of the expression of mRNA and protein of BLyS and BAFF-R,the patients had a higher level than the healthy controls (P<0.01),so did the active group than the inactive group (P<0.01).The expression levels of BLyS mRNA and protein were positively correlated to SLEDAI (P<0.01).While there was no correlation between the BAFF-R expression level and SLEDAI (P>0.05).Conclusions These findings indicate that BLyS and its receptor BAFF-R might be involved in the pathogenesis of SLE.Also,BLyS expression level might be a new parameter for the evaluation of SLE disease activity and therapeutic effect.
