Tcl/Tc2失衡和慢性光线性皮炎发病的相关性研究

Imbalance of Tc Cells in the Pathogenesis of Chronic Actinic Dermatitis

摘要: 目的 探讨Tc细胞失衡和慢性光线性皮炎(CAD)发病的相关性。方法 应用免疫组化SP法检测28例CAD患者和7例正常人对照者皮肤中真皮浸润细胞的免疫表型及白介素4、10、12,干扰素γ受体的表达。同时应用ELISA的方法检测33例CAD和32例正常人对照者血清中白介素4、10、12、干扰素γ的含量。结果 CAD患者皮损真皮内浸润的单一核细胞绝大部分表达为CD3+阳性的T细胞,阳性细胞百分数,平均灰度与CD20+阳性的B细胞相比有统计学意义(P均<0.001)。这些T细胞中大部分表达为CD8+阳性的T细胞,阳性细胞百分数和平均灰度与CD4+阳性的T细胞相比有统计学意义(P均<0.001)。Tcl细胞分泌干扰素γ,白介素12的受体在CAD患者真皮内浸润的Tc细胞表面的表达要高于Tc2细胞分泌的白介素10、4的受体表达(P均<0.001)。外周血干扰素γ、白介素12含量在CAD患者组和对照组中有统计学意义(P均<0.05)。结论 CAD发展可能和Tcl细胞的过度表达及Tc2细胞的表达受抑有一定的关系。

Abstract: Objective To investigate the association of imbalance of Tc1 and Tc2 cells with chronic actinic dermatitis (CAD). Methods Immunohistochemistry (S-P method) was used to test the subgroups of infiltrating cells and the expression of receptors of interleukin-4 (IL-4), interleukin-10 (IL-10), interleukin-12 (IL-12) and interferon-γ (IFN-γ) in these cells of the eruptions from 28 patients with CAD and 7 normal controls. The serum levels of IL-4, IL-10, IL-12 and IFN-γ from 33 patients with CAD and 32 normal controls were tested by ELISA. Results The major infiltrating cells in the eruptions of patients with CAD were T cells, which were significantly more than B cells (P<0.001). Among these T cells, the predominant was Tc cells, which were significantly more than Th cells (P<0.001). In the ease of infiltrating cells, the expression of receptors of IL-12 and IFN-γ was significantly higher than that of receptors of IL-4 and IL—10 (P<0.001). The serum levels of IL-12 and IFN-γ in patients with CAD were significantly increased than those in the control group (both P<0.05). Conclusions These findings indicate the imbalance of Tc1 and Tc2 cells probably plays an important role in the etiology and pathogenesis of CAD.
