c-Met蛋白和c-Met mRNA在地方性及艾滋病相关性Kaposi肉瘤中的表达

Expression of c-Met in Endemic Kaposi's Sarcoma and AIDS-related Kaposi's Sarcoma

摘要: 目的 探讨c-Met原癌基因蛋白和c-Met mRNA在地方性及艾滋病相关性Kaposi肉瘤不同期的表达。方法 对源自坦桑尼亚首都Muhimbili医疗中心病理科的30份标本,用免疫组化、原位杂交方法进行了研究。结果 根据Kaposi肉瘤组织中血管成分和梭形细胞的比例分为早期血管瘤样型(11个病灶)、晚期梭形细胞型(14个病灶)和中期混合型(13个病灶)。c-Met蛋白在早期表达弱,晚期表达强,中期介于两者之间,相互间差异有非常显著性(P<0.01);c-Met mRNA在不同期的表达与c-Met蛋白的表达类似。c-Met蛋白和c-Met mRNA在地方性和艾滋病相关性Kaposi肉瘤的表达差异无显著性(P>0.05)。结论 c-Met表达的改变在地方性和艾滋病相关性Kaposi肉瘤的发生发展过程中起了作用。

Abstract: Objective To study the expressions of c-Met at differential stages of endemic Kaposi's sarcoma (KS) and AIDS-related KS and its roles in the pathogenesis. Methods Twenty-one cases of endemic KS and 9 cases of AIDS-related KS from Pathological Department, Muhimbili Medical Centre, Tanzania, were analyzed clinicopathologically. Among them, 21 cases were cutaneous KS, and other 9 cases were lymph node KS. The classification was based on the histopathological characteristics of KS. Immunohisto-chemistry and hybridization in situ were performed for the detection of c-Met protein or c-Met mRNA in endemic KS and AIDS-related KS. Resuits KS were classified into three stages, i.e. early stage (angiomatous type, 11 lesions), advanced stage (spindle cell type, 14 lesions), and medial stage (mixed type, 13 lesions) according to the proportion of blood vessel components and spindle cells in the lesions. There was significant difference in the c-Met protein expression on different stages (P<0.01), with similar situation of c-Met mRNA expression. There was no difference in c-Met protein and c-Met mRNA expression between epidemic KS and AIDS-related KS (P>0.05). Conclusion The up-regulated expression of c-Met protein and c-Met mRNA in KS suggests that c-Met may play an important role in the pathogenesis and development of endemic KS and AIDS-related KS.
