增殖细胞核抗原、细胞周期素E及细胞周期蛋白依赖性激酶抑制因子p21和p27在尖锐湿疣中的表达

Expression of Cell Cycle Related Factors PCNA,CyclinE,p21(WAF1/CIP1),and p27 in Condyloma Acuminatum

摘要: 目的 探讨细胞周期相关因子增殖细胞核抗原(PCNA)、细胞周期素E(cyclinE)、p21(WAF1/CIP1)、p27在HPV6/11相关尖锐湿疣(CA)病变中的各种变化及其意义.方法 应用原位杂交技术诊断HPV6/11相关CA,采用SP免疫组化染色技术检测50例HPV6/11阳性CA患者皮损、25例正常人皮肤(包皮)中PCNA、cyclinE及p21、p27的表达及分布.结果 ①CA皮损中PCNA、cyclinE、p21的表达较正常人上皮中均有不同程度的增强,p27的表达则无统计学差异;CA皮损中其表达主要分布于棘层及颗粒层.②CA皮损中cyclinE与p21和p27的表达、p21与p27的表达间存在相关性.结论 HPV6/11阳性尖锐湿疣中,PCNA、cyclinE、p21的表达增强,并可能通过PCNA、cyclinE、p21、p27的相互协调或制约,引起自身限制的角质形成细胞增殖.

Abstract: Objective To investigate the changes of cell cycle related factors [PCNA,cyclinE,p21(WAF1/CIP1) and p27] in condyloma acuminatum(CA) and their significance.Methods HPV6/11 associated CA was diagnosed by in situ hybridization and the expression and distribution of PCNA,cyclinE,p21 and p27 in 50 HPV 6/11 positive CA specimens and 25 normal human skins (foreskins) were detected by SP immunohistochemistry technique.Results ① The expression of PCNA,cyclinE and p21 in skin lesions of CA was higher than that in normal epithelium,and there was no statistic difference for the expression of p27,and the distribution of these molecules was mainly in spinous and granular layers in CA.② There were correlations of different molecule expression in CA:between cyclinE and p21 and p27,between p21 and p27.Conclusion The data suggest that the expression of PCNA,cyclinE and p21 in HPV6/11 positive CA is increased and the coordination or restriction among PCNA,cyclinE,p21 and p27 might induce self-limited proliferation of kera-tinocytes.
