毛乳头细胞高效培养方法探索

An Efficient Method for Isolation and Cultivation of Human Scalp Dermal Papilla Cells

摘要: 显微解剖法分离毛乳头效率低,细胞生长差,采用胶原酶D消化法直接将毛囊下部的真皮鞘组织消化成真皮鞘细胞和毛乳头,从而省去了在解剖显微镜下分离毛乳头的步骤,显着减少了工作量,提高了效率,而且极大地促进了毛乳头的贴壁和细胞的生长,并保证了毛乳头的完整性,且可大批量分离出毛乳头,组织化学染色表明毛乳头细胞是一种高度特异的成纤维型细胞.

Abstract: The most commonly used method to obtain the human scalp anagen hair dermal papillae is the microdissection on excised tissue. This method have many drawbacks, such as laboriousness, low efficiency, uneasy adhesion, and poor growth. Here we report a more efficient method for the isolation of dermal papillae from human scalp hair follicles. The dermal sheath of lower part of hair follicles was entirely digested by collagenase D into isolated dermal sheath cells and dermal papillae. The advantages of the modified method are: microdissection can be omitted, work load is reduced, it is more efficient, and shows considerable improvement on the cell adhesion and growth. Futhermore, the dermal papillae can be isolated on a large scale and remained intact. The dermal papilla cells showed positive stain with alcian blue, PAS and toluidine blue, which implies that the dermal papilla cells is a highly specialized fibroblasts.
