摘要： 为了获得纯化的重组BPAG2片段,通过基因工程的方法在体外对BPAG2片段的重组cDNA克隆进行了诱导表达.重组质粒pKPL-BPAG2转化至大肠杆菌POP2136中并经SDS-PAGE分析表达产物.结果细菌表达出了分子量为22500的非融合性BPAG2片段,与对应的cDNA长度相符合,凝胶扫描分析表达产物占细菌总蛋白的18.1%.重组片段经SDS-PAGE纯化后,利用免疫印迹方法证实保留了BPAG2特异的免疫活性,我们认为该重组蛋白片段可以进一步应用于类天疱疮的体外诊断试验.

Abstract: We analysed the expression of the BPAG2 recombinant cDNA clone to obtain the purified BPAG2 fragment in vitro.The nonfused fragment of 22.5 kD was expressed by temperature induction after the pKPL-BPAG2 transformed into bacteria POP2136.This expression band was correspondent to its cDNA clone.The product accounted for 18.1% of total bact erial proteins.The recombinant fragment was purified by SDS-PAGE and ident ified that it maintained specific immunological features of BPAG2.The seresult smight be used as an in vitro diagnostic test of BP.
