树实验麻风的组织病理学观察

Histopathological Study on Experimental Leprosy in Tupaias(Tupaia Belangeri Yunalis)

摘要: 为了探讨低等灵长类动物——树作为实验麻风新模型的可能性,我们用人麻风杆菌(5.0×104~2.67×107)经静脉、前后足垫、鼻、耳皮下和睾丸内不同途径接种10只树.从感染到剖杀时间为373~786天,平均感染期为546天.组织病理学检查结果表明,在接种部位出现大片麻风性肉芽肿损害,肉芽肿内抗酸杆菌(AFB)形态指数高达40%以上.细菌密度指数为5+~6+,仅少数为3+~4+.病变部位以皮下为多见.皮神经及横纹肌为好发部位.在皮神经内外见到浸润细胞内含大量的AFB及菌团,正常神经结构受到严重破坏或呈洋葱皮样改变,近似人麻风的界线类偏瘤型病变.菌计数检查结果表明,最高增殖菌量达5.97×109/g组织.根据树所发生的组织病变及对增殖菌的鉴定证明,树实验麻风动物模型已经建立.

Abstract: Ten tupaias were infected with 5.0×104~2.67×107M.leprae by diferent routes,intravenous and subcutaneous (fore and hind foot pads,nose,ears)or intratesticular inoculation.These animals were sacrificed 373~786 days after infection,the average time were 546 days.Large patches of leprous granulomatous infiltration,containing a large number of acid fast bacili(AFB) were found at the inoculated sites by histopathological examination.The morphologic indices(MI) of most AFB were greater than 40%,and the bacterial indices(BI) ranged 5+~6+.The dermal nerves showed“onion skin-like”appearance and were severely destroyed with large numbers of AFB and globi.It looks like borderline lepromatous leprosy in tissue sections.The bacili in the foot pads of tupaia increased up to 5.97×109/g of tissue.Acording to the histopathological observation and bacteriological determination,it is proved that experimental leprosy model in tupaia has been established.
