联合用药对裸鼠麻风杆菌的影响

摘要： 为了改进麻风联合化疗方案,研究了新的抗麻风杀菌药物二甲胺四环素,氟嗪酸和氨苯砜,利福平组合的三种联合治疗方案的抗麻风杆菌效果。采用动力法投药,裸鼠在接种麻风杆菌后84～179天治疗。实验结果表明,0.01%二甲胺四环素+0.001%氨苯砜;二甲胺四环素+氨苯砜+0.05%氟嗪酸;二甲胺四环素+氨苯砜+0.01%利福平联合方案均有协同作用和明显的抗菌活性。3联优于2联。3联中以二甲胺四环素+氨苯砜+利福平方案为最佳,在停止治疗后5个月仍未发现麻风杆菌在裸鼠双后足垫中增殖。

Abstract: In order to improve multidrug treatment (MDT) regimens for leprosy patients,we stud-ied the activity of the combination of several new bacteriocidal drugs against M.leprae in the nude mice. M.leprae-infected nude mice were treated from day 84 to 179 after inoculation using kinetic method.We evaluated three regimens..minocycline(M) 0.01%+dapsone(D)0.001%;M+D+ofloxacin(O)0.05%; M+D+refampicin(R)0.01% in diet.The results indicated that there were additive or synergistic activity against M.leprae,furthermore,combination of three effective drugs were confirmed to be superior to that of two drugs.However,bacteriocidal activity of M+D+R was the best in these regimens,5 months after stopping therapy,no multiplication of M.leprae were obsorved in both hind foot pads of inoculated nude mice.
