异位性皮炎皮损处郎格罕细胞的抗CD23/FcεRⅡ反应性

Anti-FcεRⅡ/CD23 reactivity on Langerhans cells in atopic dermatitis skin lesions

摘要: 郎格罕细胞(LC)携带IgE是异位性皮炎(AD)的特征.为探索IgE如何结合至LC,检查了AD皮肤中抗CD23/FcεRⅡ反应性.26例AD皮损中18例检出抗CD23单克隆抗体MHM6反应性细胞.双重标记表明部分MHM6+细胞是CDla+LC.酸处理导致MHM6及抗IgE反应性丧失,但LC的IgE结合能力并不受影响,表明CD23/FcεRⅡ虽能从AD皮肤LC上检出,并不为LC结合馆E所必需.推测IgE通过FcεRⅡ以外的受体而结合至LC.

Abstract: Occurrence of IgE molecules qn Langerhans cells (LC) in lesional sites of atopic dermantis (AD) is a feature of this disease. The events of IgL binding on LC is unknown. In order to address this issue, we examined the presence of the low affinity IgL-Fc receptor (FcεRⅡ/CD23). In 18/26 cases, we detected cells which reacted with one particular antiCD23 mAb MHM6 in Iesional skin. Double immunoiabelling experiments revealed that a portion of MHM6+ cells was CD1a+LC. Preexposurce of AD sections to glycine-HCl (pH2.5) led to a Loss of MHM6-and anti-IgE-reactive cellular sites. In contrast, the IgE-binding capacity of LC was not affected by acid Ereatment. These observations suggest that FcεRⅡ/CD23, although detected nn LC in AD skin, is nut required for cellular IgE-binding and lead to the hypoEhesis that IgE can bind to LC via a receptor other than FcεRⅡ.
