裸鼠实验麻风动物模型的研究

ESTABLISHMENT OF THE EXPERIMENTAL LEPROSY MODEL USING IMMUNOLOGICAL DEFECT ANIMALNUDE MICE

摘要: 本文是我国首次应用免疫缺陷动物—裸小鼠(NIH/nu/nu)建立实验麻风动物模型的报告.用多菌型麻风病人体内的麻风杆菌经足垫皮下途径感染裸鼠,并用免疫正常小鼠作平行比较.结果表明,在感染早期,麻风杆菌在两种小鼠足垫中的生长曲线几乎类似,但感染后一年,菌在裸鼠足垫中增殖大大高于CFW系小鼠,前者为1.21×108/足垫,比接种时增殖12100倍,后者为4.20×106/足垫,仅增殖420倍.感染后476天,裸鼠足垫作组织病理学检查,见到瘤型麻风病变含有大量的抗酸杆菌(AFB)及许多大小菌团.菌播散到耳、鼻、坐骨神经、淋巴结、肝、脾、肺、睾丸、舌和对侧未接种足垫等部位.现已取得裸鼠体内增殖的AFB是人麻风杆菌的初步依据.

Abstract: This is the first report in China, in whichimmunological defect animal-nude mice(NIH/nu/nu) were used for the establishment of experimental leprosy model. M. leprae taken from multibacillary leprosy patient were inovulated into the right hind foot-pad of nude mice and the results were compared with that of normal mice of CFW strain. The results showed that in early stage of infection similar growth curve was observed in two strains of animals.
