Karyotype Analysis of Cryptococcus Neojormans by Pulsed-rield Gel Electrophoresls

Abstract: Chromosomes from Cryptococcus neoformans were separated by pulsed-field gel electrophoresis.Twenty strains of C.neoformans including all five serotypes and three var.species were studied.The results showed the genomes of C.neoformans are composed of 10-12 chromosomes ranging in size from 0.4-2.1 Mb,and total DNA is about 15 Mb.A good correlation was observed between the electrophoretic karyotype and the serotype of C.neoformans.It was found that each strain had a unique,reproducible pattern of chromosome bands,so this strain polymorphism can be used as an epidemiologic marker of C.neoformans infection.
