Determination of DNA-fluorescence Intensity of Candida Species with Adherent Cell Analysis and Sorting Cytometer

Abstract: DNA-fluorescence intensity of Candida isolates including C.albicans, C.tropicalis and C.guilliermondi growing in exponential -growth-phase was determined by adherent cell analysis and sorting cytometer.All specimens were treated with RNAase and stained with propidium idodide(PI).The results showed that interspecific difference in average fluorescence value of exponential-growth-phase was found, the average flourescence value of C.tropicalis was the highest, and that of C.guilliermondi was the lowest.It is suggested that the average fluorescence value, the intensity of DNA fluorescence particle in single cell is a stable and interspecific parameter, which may be used as an useful method for the classification of fungus.
