Analysis of T-cell receptorγgene rearrangement for cutaneous T-cell lymphomas

Abstract: Objective To study the early diagnosis of cutaneous T-cell lymphoma(CTCL).Methods DNA from39formalin-fixed,paraffin-embedded tissues of 24 cases of CTCL,2 cases of dubious lymphoma,and4cases with non-specific erythroderma,was amplified by polymerase chain reaction(PCR),with special primers of TCR Vγ1-8,Vγ9,and Jγ1/γ2 to analyze T-Cell receptorγgene clonal rearrangement(TCR-GR clonal).The specimens from30patients with non-specific dermatitis were taken as negative control.Results Clonal of TCR Vγ1-8 was demonstrated in38of39paraffin-embedded specimens.Vγ9-GR was shown in 37 of 39 specimens TCRγ-GR was shown especially in 13 patients with MF of early stage,2 patients with dubious CTCL,and4patients with non-specific erythroderma,as well as in 6 of 30 patients with histiologically nonspecific dermatitis which suggested that these 6 cases were "clonal dermatitis".Conclusion Clonal TCR γ-GR may be detected in patients with early CTCL,even when the histologic findings are not unequivocally diagnostic.
