IL-1αmRNA Expression and Protein Secretion After Ultraviolet Irradiation in Human Keratinocyte Originated SCC 12F Cells

Abstract: Objective To investigate IL-1α mRNA expression and protein secretion after ultraviolet irradiation in human keratinocyte(KC)originated SCC 12F cells.Methods Expression of IL-1α mRNA was detected by Northern blot and the secretion level of IL-1α protein was analyzed by ELISA.Results There was a spontaneous expression of IL-1α mRNA in SCC 12F cells in culture system of normal KC,which was increased along with the culture time,and reached the highest expression level in 120 hours.The secretion level of IL-1α protein was increased in a time-and dose-dependent manner in SCC 12F cells after ultraviolet B(UVB)irradiation.Conclusions SCC 12F cells may express IL-1α mRNA spontaneously under regular KC media.The secretion level of IL-1α protein shows both time-and dose-effect after UVB irradiation.
