Action mechanism of narrow-band UVB on human umbilical vein endothelial cells

Abstract:

Objective: To investigate the biological activity and the related cytokines of vascular endothelial cells treated with the narrow-band ultraviolet B(NB-UVB), and to evaluate the mechanism of action of NB-UVB on the vascular endothelial cells. Methods: The cells of human umbililcal vein endothelial cells (HUVEC) were incubated for 24 hours after the cells were treated with different dosage (50、100、200、300mJ/cm2) of NB-UVB radiation.Cell proliferation rates were determined by the MTT assay. The VEGF、MMP-1 and MMP-2 were determined by ELISA. Results: Morphological damage to different degrees was observed in all groups, including cytomorphosis, hydropic degeneration and the increase of cell debris. The changes was dependent on the dose of NB-UVB. The results of MTT assay showed that a significant decrease was observed, and the changes were dose-dependent. The secretion of VEGF and MMP-1 was significantly decreased. The secretion of MMP-2 was no difference except the group of 300mJ/cm2. Conclusion:.NB-UVB treatment caused a stable decrease of cell proliferation rates, and the changes were dose-dependent. NB-UVB treatment can inhibit the secretion of VEGF and MMP-1, but MMP-2.
