Study on the ORF75 subtypes of human herpesvirus–8 in Xinjiang patients with Kaposi’s sarcoma

Abstract:

Objective: To study the epidemic situation and characteristic of ORF75 subtypes of Human herpesvirus-8 in Kaposi′ s Sarcoma in Xinjiang, and to primary discuss the correlations between different forms of Kaposi’s sarcoma and different subtypes. Methods: HHV-8 DNA were amplified by nested-PCR and sequenced which was extracteded from 25 cases paraffin-embedded tissue from the Xinjiang .The phylogenetic analysis were carried out by using the software DNASTAR, the program Clustal W and the PHYLIP package and then the ORF75 subtype was confirmed . Correlations between different clinics of Kaposi’s sarcoma and different subtypes were determined by Fisher’s exact probabilities. Results: ①HHV-8 DNA was detected in 25(84%) out of 21 cases and 7 cases AIDS-KS (100%) in Xinjiang . ②Based on the partial ORF75 nucleotide sequence analysis, indicate that Eighteen of these strains belong to the A subtype , with three belonging to the C group.③The different ORF75 subtypes of HHV-8 among mucosa damage and clinical classification of Kaposi′ s Sarcoma were not significant (P >0.05).Conclusion:①ORF75 gene is detected as epidemiologic study of HHV-8. ②The majority of Kaposi′ s Sarcoma patients in Xinjiang were infected by the A and C subtype of HHV-8. ③The different ORF75 subtypes of HHV-8 have nothing to do with mucosa damage and clinical classification of Kaposi′ s Sarcoma.
