Affection of MTX on the expression of TLR2、TLR4 in human peripheral blood mononuclear cell of patients with psoriasis vulgaris

Abstract:

【Abstract】Objective To investigate the regulation of MTX on the expression of TLR2、TLR4 in human peripheral blood mononuclear cell of patients with psoriasis vulgaris and explore the treatment mechanism. Methods The Positive rates of TLR2 and TLR4 in human leucocytes were measured by flow cytometry in patients with psoriasis vulgaris before and after treatment with MTX. Results The Positive rates of TLR2 and TLR4 in human leucocytes were significant increase in patients with psoriasis vulgaris as compared with normal controls (P<0.01), and there was significant difference between the active stage of psoriasis vulgaris and the resting stage(P<0.05). There was significant difference between before and after treatment with MTX (P<0.01). The expression of TLR2 and TLR4 was not correlated with PASI scores(P>0.05). Conclusions The TLR2 and TLR4 and the innate immune response mediated by it may be very important to the pathogenesis of psoriasis vulgaris. It is indicated that inhibiting the expression of TLR2、TLR4 may be involved in the treatment of psoriasis vulgaris with MTX.
