Expression of HINT/PKCI-1 mRNA in patients with malignant melanoma and nevus

摘要： 目的 探讨组氨酸三聚体核苷结合蛋白1(HINT1)mRNA在恶性黑素瘤和痣细胞痣中的表达.方法 采用半定量RT-PCR方法检测了25例痣细胞痣和25例恶性黑素瘤中HINT1mRNA的表达,包括新鲜标本和存档石蜡标本.结果 相同标本经过10%甲醛固定、石蜡包埋后提取RNA进行半定量RT-PCR的结果与新鲜冻存组织提取RNA的结果高度一致,痣细胞痣组织中HINT1mRNA的表达水平为0.49±0.04,恶性黑素瘤组织中HINT1mRNA的表达水平下降为0.31±0.07,差异有统计学意义(P<0.05).结论 HINT1mRNA表达下降可能与恶性黑素瘤发病有关.

Abstract: Objective To investigate the expression of HINT1 mRNA in the tissue of malignant melanoma and nevocellular nevus. Methods Half-quantitative RT-PCR was used to measure the expression of HINT1/PKCI-1 mRNA in fresh and archival tissues of 25 patients with malignant melanoma and 25 patients with nevus. Results In the case of the same specimen, the expression of HINT/PKCI-1 mRNA showed no difference in the tissue treated with formalin fixation and paraffin embedment with that in the fresh tissue. The expression level of HINT/PKCI1 mRNA was 0.49±0.04 in nevus and 0.31±0.07 in malignant melanoma, and the difference was significant (P<0.05). Conclusion The Results indicate that the reduced expression of HINT1 mRNA might be related to the pathogenesis of malignant melanoma.
