Effects of Arsenic Trioxide on the Tyrosinase Activity and Melanogenesis in Human Malignant Melanoma Cell Line A375

摘要: 目的 探讨三氧化二砷(As2O3)对A375人恶性黑素瘤细胞株酪氨酸酶活性及黑素生成的影响。方法 利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,采用酶学方法测定酪氨酸酶活性,465nm比色法测定黑素含量。结果 As2O3浓度<0.01μmol/L时,随As2O3浓度的增加,促进黑素瘤细胞的增殖、黑素的合成及上调酪氨酸酶的活性;As2O3浓度等于0.01μmol/L时作用最强。0.1~1μmol/L则表现为抑制作用;>1μmol/L则出现细胞毒性作用。结论 经As2O3处理后A375人恶性黑素瘤细胞株的细胞增殖、酪氨酸酶活性及黑素的生成与As2O3浓度呈双相性变化。

Abstract: Objective To study the effects of arsenic trioxide on the tyrosinase activity and melanogenesis in human malignant melanoma cell line A375. Methods MTT was used to measure the proliferation of human malignant melanoma cell A375. The tyrosinase activity and the melanin content were measured. Results When the concentration of arsenic trioxide was lower than 0.01 μmol/L, a positive effect was shown along with the increase of the concentration, such as cell proliferation, tyrosinase activity and up-regulation of melanogenesis. When the concentration of arsenic trioxide was above 0.01 μmol/L, an inhibitory effect was observed. If the concentration of arsenic trioxide was above 1μmol/L, cytotoxic effect occurred. Conclusion Arsenic trioxide has diphasic effect on human malignant melanoma cell line A375.
