培养人角朊细胞中白介素15的基因表达

摘要: 目的 为了探讨白介素15(IL-15)在表皮角朊细胞中的表达及调节.方法 应用RT-PCR及Northern杂交方法分析了IL-15在培养的人角朊细胞及鳞状细胞癌细胞系(HSC-5)中的表达,并观察了地塞米松对角朊细胞表达IL-15的影响.结果 正常角朊细胞及鳞状细胞癌细胞系细胞均有IL-15 mRNA的表达,10-6mol/L地塞米松处理培养的角朊细胞后,IL-15 mRNA表达水平明显降低.结论 表皮细胞来源的IL-15可能介入某些炎症性皮肤病的发病过程.

Abstract: Objective To study the expression and regulation of interleukin-15 (IL-15) in epidermal keratinocytes. Methods The expression of IL-15 mRNA in cultured normal human keratinocytes (NHKs) and human squamous cell carcinoma cell line (HSC-5) was analysed, and the effect of dexamethasone on IL-15 mRNA expression was studied by using RT-PCR and Northern blotting technique. Results The results showed that both NHKs and HSC-5 expressed IL-15 constitutively. The level of IL-15 mRNA was significantly decreased after the cells were cultured with 10-6M dexamethasone. Conclusion It is suggested that keratinocyte-derived IL-15 might be involved in the development of certain inflammatory skin diseases.
