白介素18和干扰素γ在生殖器疱疹感染中的意义

Role for Interleukin-18 and Interferon-gamma in Genital Herpes Simplex Virus Infection

摘要: 目的 探讨白介素18(IL-18)和干扰素γ(IFN-γ)在单纯疱疹病毒2型(HSV-2)感染的生殖器疱疹患者发病机制中的意义。方法 采用双夹心ELISA方法对43例确诊为HSV-2感染的生殖器疱疹患者外周血单一核细胞(PBMC)及血清IL-18和IFN-γ进行检测,设立健康对照组。结果 HSV-2感染的生殖器疱疹患者的血清及PBMC中IL-18水平分别为(424.32±51.60)ng/L和(417.54±47.23)ng/L,均明显高于对照组,差异有显著性(P<0.01);HSV-2感染者血清IFN-γ水平为(384.84±72.45)ng/L,明显高于对照组,两组差异有显著性(P<0.01);PBMCIFN-γ水平(381.62±70.45)ng/L,亦明显高于对照组,差异有显著性(P<0.01);HSV-2感染生殖器疱疹患者血清IL-18和IFN-γ呈正相关(r=0.515,P<0.05);rhIL-18可诱导PBMC产生IFN-γ。结论 HSV-2感染的生殖器疱疹患者体内IL-18水平升高,并与IFN-γ呈正相关,rhIL-18可刺激HSV-2感染者PBMC产生IFN-γ,IL-18具有明显的抗HSV-2感染作用,其机制可能与诱导IFN-γ产生有关。

Abstract: Objective To study the roles of interleukin-18 (IL-18) and interferon-gamma (IFN-γ) in the pathogenesis of infection with herpes simplex virus type 2 (HSV-2) in patients with genital herpes. Methods Double-sandwich ELISA was used to detect the levels of IL-18 and IFN-γ in PBMCs and sera of 43 patients with genital herpes who were confirmed to be infected with HSV-2, and healthy volunteers as control group. Results IL-18 levels in the sera and PBMCs in the patients with genital HSV-2 infection were (424.32±51.60)ng/L and (417.54±47.23)ng/L, respectively, and were significantly higher than those in the control group (P<0.01). IFN-γ levels in the sera and PBMCs in HSV-2 infected patients were (384.84±72.45)ng/L and (381.62±70.45)ng/L, respectively, and significantly higher than those in the control group (P<0.01). A positive correlation between IL-18 and IFN-γ levels was found in the patients (r=0.515, P<0.05). IFN-γ could be induced by rhIL-18. Conclusion An increased level of IL-18 is found in the patients with genital HSV-2 infection and also correlated with increased level of IFN-γ. The rhIL-18 can stimulate PBMC in HSV-2 infected patients to produce IFN-γ, and IL-18 has a significant role against HSV-2 infection, which may be related to the production of IFN-γ.
