干细胞标志物CD133、nestin和CD44在恶性黑素瘤皮损中的表达及意义

Expression of stem cell markers CD133、Nestin and CD44 in malignant melanoma and its significance

摘要:

目的:探讨干细胞标志物CD133、Nestin和CD44在恶性黑素瘤中的表达及意义。方法: 采用免疫组化SP法检测3种标记物在30例恶性黑素瘤及30例皮内痣皮损中的表达。结果: ⑴ CD133、Nestin和CD44在恶性黑素瘤组织中的阳性表达率分别是53.33%,80.00%和30.00%,在皮内痣分别是23.33%,53.33%和0,各标志物组间差异比较均有统计学意义(P<0.05);⑵ CD133、Nestin和CD44在恶性黑素瘤组织中阳性细胞率平均值分别为2.98%±5.62%,34.92%±34.89%和1.28%±3.26%,在皮内痣分别为0.10%±0.21%,7.26%±13.13%和0,各标志物组间差异比较均有统计学意义(P<0.05);⑶各组中阳性细胞率与患者性别、年龄、病程比较,差异无统计学意义(P>0.05)。结论:干细胞标志物CD133、Nestin和CD44在恶性黑素瘤中高表达,在皮内痣中低表达或不表达,这提示恶性黑素瘤中可能存在肿瘤干细胞。

Abstract:

Objective:To investigate the expression of stem cell markers CD133, Nestin and CD44 in malignant melanoma and its significance. Methods: The expression of three markers in malignant melanoma and intradermal nevus tissues were immunohistochemically detected. Results: ⑴In malignant melanoma tissue, the positive expression rate of CD133, Nestin and CD44 were 53.33%, 80.00% and 30.00% respectively, while in intradermal nevus they were 23.33%, 53.33% and 0 respectively.The difference of each marker between malignant melanoma and intradermal nevus was of statistically significant (P<0.05). ⑵The percentage of positive cells of CD133, Nestin and CD44 were 2.98%±5.62%,34.92%±34.89% and 1.28%±3.26% respectively,while in intradermal nevus they were 0.10%±0.21%,7.26%±13.13% and 0 respectively. The difference of each marker between malignant melanoma and intradermal nevus was of statistically significant (P<0.05). ⑶No correlation was found in each group between the percentage of positive cells and sex ,age and the course of disease (P>0.05). Conclusions: The positive expression rate of CD133, Nestin and CD44 was higher in malignant melanoma than that in intradermal nevus. it has been suggested that cancer stem cells might exist in malignant melanoma.
